Artane (trihexyphenidyl) pharmacologic effect

Artane (trihexyphenidyl) is an antispasmodic drug for the treatment of Parkinson's disease. It effectively treats extrapyramidal symptoms associated with treatment with neuroleptics (torsion dystonia, orofacial dyskinesia, Oculogyric crisis, akathisia).

Artane has inflammatory properties. It inhibits parasympathetic nervous system activity and relaxes smooth muscles.

Artane (trihexyphenidyl) indications

Artane (trihexyphenidyl) is used to treat Parkinson's disease, all forms of parkinsonism and other lesions in the extrapyramidal tract: spastic paralysis of central origin, and others.

Artane is also used for the treatment of depression syndrome and reduced mental capacity.

Artane does not contribute to the development of glaucoma. Therefore, it is particularly effective for the treatment of atherosclerotic parkinsonism.

Artane (trihexyphenidyl) dosage and administration

Artane (trihexyphenidyl) is taken during or after a meal, starting with a dose of 1 mg per day. The dosage may be increased to 2-4 mg (in 3-4 divided doses) depending on individual tolerability. It is desirable to take Artane with food (to avoid nausea).

Patients receiving other medicines should slightly reduce the dose of other medications.

The highest single dose of Artane is 10 mg. The highest daily dose is 20 mg.

What are the side effects for Artane (trihexyphenidyl)?

Adverse events (confusion, anxiety) are usually observed in patients with atherosclerotic parkinsonism. They should begin treatment with very low doses and slowly increase in dose as therapy continues.

Nervous system: headache, dizziness, irritability, delusions, hallucinations, fatigue, decreased ability to concentrate, psychosis.

Digestive system: nausea, vomiting. Allergic reactions: skin rash. Other reactions: dryness of the oral mucosa, paresis of accommodation, high intraocular pressure, constipation, urinary retention, tachycardia, sweating, decreased salivation, flushing, drug dependence.

What should I know about Artane (trihexyphenidyl) contraindications?

Artane (trihexyphenidyl) contraindications: hypersensitivity to any component of the drug, nonmalignant stenosis of the gastrointestinal tract, angle-closure glaucoma, urinary retention, prostatic hyperplasia, pulmonary edema, dementia, children's age (up to 12 years), pregnancy, lactation. Artane precautions: decompensated heart failure, kidney or liver disease, tachycardia, tachyarrhythmia, hypertension, psychosis, acute myocardial infarction.

Artane (trihexyphenidyl) overdose

Large doses of the drug may cause mydriasis, dry mouth, tachycardia, fever, disorders of the central nervous system (mental and motor excitation, coma, paralysis of the respiratory center). Treatment: there is no specific antidote. Treatment is symptomatic and supportive.

Artane (trihexyphenidyl) interaction with other drugs

Artane (trihexyphenidyl) should not be used with H1 histamine receptor blockers, phenothiazine derivatives, tricyclic antidepressants.

Antiparkinsonian effect of trihexyphenidyl is reduced under the influence of reserpine.

Levodopa increases the effect of trihexyphenidyl. When used together with levodopa, the dosage of both drugs should be reduced. In this case, 3-6 mg of trihexyphenidyl per day is enough (divided into two doses).

Monoamine oxidase inhibitors may increase the anticholinergic effect of trihexyphenidyl.

Artane may reduce the effect of sublingual nitrates (due to dry mouth).

Depressive effects of alcohol and drugs that affect the central nervous system may be enhanced when used with trihexyphenidyl.

Artane (trihexyphenidyl) special instructions

Artane (trihexyphenidyl) should be used with caution in hypertension, liver and kidney diseases.

During the period of treatment, patients should refrain from driving and performing potentially hazardous activities.

Patients older than 60 years should use Artane with extreme caution (high risk of sensitivity to the drug).

Stopping your medication abruptly may adversely affect your condition. Trihexyphenidyl dose should be reduced gradually.

I am 49 years old and I’m suffering from benign essential blepharospasm as well as from cervical dystonia. My situation was so severe that I was very close to go blind due to those severe spasms that I was having. I have tried a lot of medicines in order to control them but nothing seemed to ever help me. only later I have finally found out about artane and honestly I didn’t thought that it would make a difference, however, I was wrong and I am so happy that I was. I started to take it approximately 7 or 8 years ago and this was and still is the drug that changed so much in my life as it greatly helped my ability to see again. since I started to take it I could control the spasms a lot better and that’s how I could go back to my work that I thought I would never be able again. I can say that this drug saved my seeing ability which results in saving my work and hence it saved my life, the quality of my life was greatly influenced by it. besides this, the drug has helped me to have a lot more energy than in the past so I was able to do a lot more things. anyway, not everything is perfect so as any other medicine has side effects this one has too and they were: constipation, dry mouth symptom and some muscle stiffness too. Besides I have also noticed that I’m having some troubles with the short term memory but I cannot tell you for sure if the drug has something to do with it. I also need to tell you that my tolerance level to this drug was quite lower than the dosage that was recommended to me. anyway, even with these side effects it is still worth using it, for me the benefits definitely outweigh the side effects as it allows me to function a lot better especially if comparing to how something else made me feel. For me this drug has improved the quality of my life, a lot so that is why I can recommend it to everybody who is suffering from dystonia who is having troubles with antro or retrocolis or with benign essential blepharospasm (at least you should give it a try). Hope it will help you the same way as it helped me.
